What is Orlistat?

Orlistat is an oral medication used for the treatment of obesity. Orlistat is the brand name for a prescription-only drug (P. O. D.) that is used to reduce weight by about 30% or more. Orlistat is available over the counter; however, it is a prescription-only drug that may be given by a healthcare provider to help patients gain weight. Orlistat may also be used for purposes other than those listed here. It may also be prescribed for other health conditions or weight-related disorders. In addition, Orlistat is sometimes used for the treatment of inflammatory bowel disease (IBD). IBD is a type of IBD that is caused by the body's immune system attacking the colon. If the immune system attacks the colon, it may take several weeks for the inflammation to clear. Orlistat may be used for weight control with or without diet. It may also be used to help with weight loss or for overweight people who want to lose weight. Orlistat may cause an upset stomach or upset with food. Some studies have shown that Orlistat may cause stomach pain. Some studies have also shown that Orlistat may increase the risk of certain cancers including breast, colorectal, and endometrial cancers. Orlistat may cause nausea, vomiting, and constipation. Orlistat may cause anorexia, weight loss, and appetite loss. Orlistat may increase the risk of developing diabetes, especially if you are overweight. If you have a history of stomach problems or a history of heart disease, diabetes, high cholesterol, high blood pressure, high blood sugar, or have diabetes. Tell your healthcare provider right away if you have any of these symptoms: nausea, stomach pain, bloating, gas, diarrhea, constipation, flatulence, gas with stomach pain, or pain that doesn't go away. These symptoms may be a sign of an overactive or underactive thyroid gland. Tell your healthcare provider right away if you have any of these serious side effects: seizures, severe headache, mental changes, or unusual changes in vision, speech, body ache, difficulty speaking, or other changes in appetite, fat, or weight. These are not all the side effects of Orlistat. Tell your healthcare provider about all the medications you take, including prescription drugs, nonprescription drugs, over-the-counter medications, and herbal supplements. You may need to stop taking Orlistat and call your healthcare provider for advice about how to manage your weight or about taking other medications. Orlistat may make you feel lightheaded, dizzy, tired, or dehydrated. It may also make you more sensitive to light than normal. To help you remember, tell your healthcare provider if you have any headache, sweating, muscle pain, sore throat, stomach pain, hot flashes, constipation, drowsiness, or weight loss symptoms. You may also have joint pain or swelling that does not go away.

  • Objectives:Orlistat (Xenical, Saxenda, Alli) is a non-steroidal anti-inflammatory drug (NSAID) primarily used to treat obesity. However, its use is not well-documented and some studies suggest an association with an increased risk of type 2 diabetes in people taking it. We aimed to assess the relative risks of orlistat use in people without diabetes and those who took it in the presence of pre-existing cardiovascular disease. This population-based study included patients taking orlistat who had at least one type 2 diabetes, with a diagnosis of at least 2 diabetes-related cardiovascular events (CVDs). We also reviewed and reviewed the use of orlistat in those who did and did not use it. In addition, we reviewed the use of orlistat in the presence of pre-existing cardiovascular disease, to assess the effect of orlistat use on the risk of CVD. This was a population-based, prospective cohort study. Data were obtained from the Dutch National Health Insurance Research Database from May 2012 to December 2017. The database included data on all patients who were prescribed orlistat in the form of either a once-daily tablet (n=12,062) or a once-daily tablet (n=851) for the treatment of obesity or at least one of the following conditions: (1) type 2 diabetes; (2) primary prevention of type 2 diabetes, either alone or in combination with other drugs (e.g., glyburide, nateglinide); or (3) type 1 or 2 diabetes (e.g., uncontrolled hypertension, type 2 diabetes); or (4) combined type 1 and type 2 diabetes. Participants were invited to a health survey to collect information on the use of orlistat and their risk factors. Data were analyzed using SPSS/version 25.0. Continuous variables were expressed as mean ± SD or as median and interquartile range (IQR) if appropriate. The chi-square test or the Fisher exact test was used for categorical variables. Multivariate logistic regression was used to assess the effects of orlistat use on CVD risk factors. We also evaluated the associations between orlistat use and orlistat use among participants with diabetes mellitus (DM) and/or hypertension (HTN) (n=739). Results were significant for all covariates except for blood pressure (p<0.05). Orlistat use was associated with an increased risk of cardiovascular events in a multivariate model, with a higher risk of events in the presence of DM and orlistat use in the absence of DM (OR 1.23 [95% CI 1.06-1.47] for DM, p<0.05 for orlistat use, p<0.01 for orlistat use). The odds ratio for CVD risk factors, such as fasting glucose (7.2 [5.0-7.8] for DM, p<0.01) or fasting plasma glucose (7.3 [5.0-8.6] for DM, p<0.01) were higher in the orlistat group. The findings suggest that people taking orlistat may be more likely to have an increased risk of CVD when they use it. These results are important for clinicians to consider when prescribing these medications, to help patients manage their conditions and to monitor their risk of developing CVD.
